Product Detail GTPases are involved in nucleocytoplasmic transport, participating in the import and export of proteins and RNA from the nucleus. They switch between the cytoplasmic GDP-bound state and the nuclear GTP-bound state through nucleotide exchange and GTP hydrolysis. Nuclear import receptors such as importin β bind their substrates only in the absence of GTP-bound RAN and release them upon direct interaction with GTP-bound RAN, whereas export receptors behave in the opposite manner. Therefore, RAN controls the cargo loading and release of transport receptors in appropriate compartments and ensures the directionality of transport. The interaction with RANBP1 can cause a conformational change in the complex formed by XPO1 and RAN, thereby triggering the release of the nuclear export signal of cargo proteins. RAN (GTP-bound form) triggers microtubule assembly on mitotic chromosomes and is essential for normal mitotic spindle assembly and chromosome segregation. It is necessary for the normal progression of mitosis.
