Background: IL7 (Interleukin-7) was first identified as a factor promoting the growth of mouse B cell precursors in bone marrow culture in 1988. It is a cytokine produced by non-lymphoid cells in the lymphoid organs. In humans, it is also produced by keratinocytes, peripheral blood dendritic cells, follicular dendritic cells, intestinal epithelial cells, liver tissue, endothelial cells, smooth muscle cells, and fibroblasts. The IL7 gene is localized to human chromosomes 8q12-q13. Is a glycoprotein with a hydrophobic core of four A-helices. Recombinant Human IL-7 is a 17.4kDa protein containing 153 amino acid residues.

Biological activity:

Interleukin 7, also known as IL7, IL-7, is a hematopoietic growth factor secreted by stromal cells in the bone marrow and thymus. It is also produced by keratinocytes, dendritic cells, hepatocytes, neurons, and epithelial cells, but not by lymphocytes. IL-7 stimulates the differentiation of pluripotent stem cells into lymphoprogenitors and also stimulates the proliferation of all cells in the lymphoid line (B cells, T cells, and NK cells). It is important for B cell maturation, T cell and NK cell survival, development, and proliferation at certain stages of homeostasis. Interleukin-7 is an important cytokine in B cell and T cell development. This cytokine and hepatocyte growth factor (HGF) form a heterodimer that acts as a pro-b cell growth stimulator. IL-7 binds to the IL-7 receptor, a heterodimer composed of the interleukin-7 receptor alpha and the common gamma chain receptor. Interleukin-7 promotes blood diseases (acute lymphoblastic leukemia, T-cell lymphoma). Elevated levels of IL-7 have also been detected in the plasma of HIV-infected patients. IL-7 has been tested as an immunotherapy agent in many preclinical animal studies and more recently in human clinical trials against various malignancies and HIV infections. Interleukin-7 also helps improve immune recovery after allograft stem cells.
