This is because the density between calcium and paraffin is different, it is difficult to cut a complete section. It is better to fix the calcium containing tissue before decalcification or both operate at the same time. Then continue next operations such as dehydration, transparency, wax immersion, embedding and slicing.There are many reagents used for decalcification; decalcification agents include organic acids, inorganic acids, ethylenediaminetetraacetic acid (EDTA), and electrolytic decalcification.

Citrate decalcification solution is a buffered decalcification solution, which is a slow decalcification solution. Its advantages are: ① good staining results of tissues decalcified by citrate decalcification solution; ② little damage to the structure of the tissue. The disadvantages are: ① decalcification speed is very slow, not suitable for routine specimen decalcification; ② decalcification of tissue will be slightly hardened. This reagent is only used in the field of scientific research, and should not be used for clinical diagnosis or other purposes.
