Alexander's stain stains fertile pollen cysts with a purplish-red color, stunted pollen cysts are smaller or lighter in color, and abortive pollen has a greenish tint. If pollen viability staining is required, TTC staining or germination assay is recommended.

Alexander's Stain pH is acidic and is a good stain for plant pollen grains.
